بررسی رونوشت های متغیر ژن BRCA1 در تومورهای پستان

Persian Abstract

مقدمه و هدف : سرطان پستان شایعترین سرطان در میان زنان می باشد ، و تقریبا تمام موارد سرطان در زنان را شامل می شود . شواهد زیادی وجود دارد که سرطان پستان نتیجه تغییرات ژنتیکی و اپی ژنتیک متعدد روی انکوژن ها، ژن های سرکوب کننده تومور و ژن های ترمیم کننده DNA است . این مطالعه ، بررسی رونوشت های متغییر ژن BRCA1 به عنوان ژن سرکوب کننده تومور در بیماران مبتلا به سرطان پستان با هدف دستیابی به یک فاکتور پیش آگهی جهت تشخیص زودرس تومور پستان است . مواد و روش ها : 50 نمونه تومور از بیماران مراجعه کننده به بیمارستان دی تهران که مبتلا به سرطان پستان بودند، از طریق جراحی تهیه شد و پس از استخراج RNA از تومورهای بیماران و سنتز First strand cDNA ، توسط پرایمرهای طراحی شده ، با روش Nested RT-PCR به تکثیر cDNA پرداخته و پس از انجام الکتروفورز رونوشت های متفاوت از رونوشت با طول کامل تمیز گردید . نتایج : پس از بررسی رونوشت های متغیر ژن BRCA1 در نمونه تومور افراد مبتلا مشخص گردید که ارتباط معنی دار بین مرحله بیماری و نیز درگیری غدد لنفاوی با وجود حذف قطعه b از اگزون 11 وجود دارد . در بررسی سایر داده ها هیچگونه ارتباط آماری معنی داری بین سایر داده های پاتولوژیکی شامل گیرنده استروژن و پروژسترون ، درجه تمایز توده و نوع آسیب شناختی با وجود پیرایش ژن BRCA1 وجود نداشت . نتیجه گیری : با توجه به این آزمایش ، می توان از داده های پیرایش متغیر این ژن در کنار اطلاعات پاتولوژیکی برای تعیین وضعیت بدخیمی تومور و نیز در صورت انجام مطالعات بیشتر ، برای پیش بینی میزان بقای بیماران بهره برداری نمود.

Alternative Splicing of BRCA1 in Breast Tumors

English Abstract

Background and objective: Breast cancer is the most pervasive cancer among women. About 33 parcent of cancers in women is the Breast cancers. Various evidences indicate that Breast cancer drive from various genetic and epigenetic alternations of oncogenes, tumor suppressor genes and DNA repair genes. This study has been focused on the investigating Alternative splicing of BRCA1 genes as a tumor suppressor in the Breast tumor patient for approaching a prognostic value. Materials and methods: 50 samples of tumor pations has been prepared through surgery prosegers with Breast concer. They were invited to Day hospital in Tehran . Then RNA were extracted from patient tumors, and first strand cDNA were synthesized Then through designed primers, cDNA had been generated through nested RT-PCR. After electrophoresis different splicing from the splicing with complete length were separated . Result: After verifying the alternative splicing gene of BRCA1 in the tumor sample of patient in has been recognized that there is an acceptable coordination between the stage of cancer and involved lynph node with the omitted piece of b from the exon 11. in the study other data there is no meaningful statistics between other pathologic data that include the receptor of Estrogen and progesterone, grade of tumor and the type of pathology. Discussion: According to this study, we can use alternative splicing data of this gene beside the pathological information for specifying the malignancy situation of tumor. So we can predispose the survival of the patients with more studies.
