Title

physical exercise increases the number of endogenous neural stem cells in aged mice

English Abstract

While it has recently been demonstrated that physical activity can reverse the age-related decline in hippocampal neurogenesis, little is known regarding the effect of running on endogenous neural stem cells (NSCs). Accordingly, we sought to: a) determine the prevalence, distribution, and age-related decline of NSCs lining the lateral ventricles and b) determine whether increased physical exercise modulates NSC numbers in ageing mice. As the majority of neurospheres are derived from progenitor cells, in addition to the neurosphere assay we employed the recently developed neural-colony forming cell assay (N-CFCA) to determine the prevalence of NSCs. Thus, we serially sectioned (400 um) the brains of juvenile (6-8 weeks), 6, 12, 18, and 24-month old mice, micro-dissected the PVR from each slice, and cultured equal numbers of cells in the neurosphere assay and N-CFCA. A second cohort of 12-month old mice were given access to a running wheel for 10 or 21 days prior to sacrifice to determine the effects of running on the NSCs. While sphere forming cells (SFCs) and NSCs were detected in naïve mice throughout the ventricular neuraxis at all ages, the extent of the age-related decline varied depending upon the location of NSCs along the neuraxis. At all ages investigated a decline in the prevalence of NSCs was observed including a maximal reduction of 88.5±1.3% (Juvenile vs. 24-month). Of interest, the number of NSCs detected in 12-month old runners (21 days) was significantly higher relative to age matched naïve mice (79±5.2 versus 33.6±0.3,p<0.001), demonstrating for the first time that exercise induces the proliferation of NSCs and providing proof of principle that environmental factors can increase the number of NSCs in at least one region of the adult brain.
