Interleukin-18 gene promoter polymorphisms and recurrent spontaneous abortion

English Abstract

Recurrent Spontaneous Abortion(RSA) is one of the immunological complications of pregnancy. However,the etiology of RSA is still unknown.It has been accepted that women with normal pregnancy have a higher Th2 cytokine profile,while women with a history of RSA tend to produce Th1 type response.IL-18 formally known as IFN-g inducing factor seems to participate in the regulation of Th1/Th2 response . The aim of our study was to evaluate the frequency of two Single-Nucleotide Polymorphisms (SNPs) at the IL-18 promoter region in patients with RSA in comparison with healthy pregnant control subjects.DNA from peripheral blood of 102 patients with RSA and 103 apparently healthy pregnant subjects were evaluated for changes in promoter of IL-18 gene (position 137G/C and position 607C/A) by Allele-Specific-PCR(AS-PCR) technique. The frequency of each allele was compared in patient and control groups.Results indicated that the frequencies of GG,CC and GC at position 137 in patients were 55.89%,4.9% and 39.21%respectively. These figures in controls were revealed to be 54.37%,7.77% and 37.86%(P=o.7).Similar analysis for position 607 revealed the frequency of CC,AA,CA in patients as 36.27%,23%,41.18% and for controls 31.07%,16.5% and 52.43% respectively(p=0.25). Results of this investigation indicated that there were no significant difference in frequency of these SNPs in RSA and healthy normal subjects.
