مقدمه: 5-فلوئورواوراسیل، داروی اصلی مورد استفاده در سرطان کولورکتال است که به دلیل اثر غیراختصاصی بر سلولهای سالم، با عوارض جانبی متعددی همراه میباشد. یکی از راهکارهای فائق آمدن بر اثرات جانبی نامطلوب دارو، بارگذاری دارو در نانوذرات هدفمند است. در مطالعهی حاضر، نانوذرات اکسید آهن (Fe3O4) با پوشش کوپلیمری پلیلاکتیک اسید-هیالورونیک اسید (PLA-HA)، جهت بارگذاری داروی 5-فلوئورواوراسیل سنتز گردیده و خصوصیات فیزیکوشیمیایی، الگوی رهش دارو از نانوذرات، سمیت و کارآیی آنها در انتقال دارو به ردهی سلولهای سرطانی کولورکتال (HCT116) مورد ارزیابی قرار گرفت. روش کار: نانوذرات Fe3O4 به روش سنتز سبز تهیه شده و جهت تایید سنتز نانوذرات اکسید آهن از طیف بینی مادون قرمز تبدیل فوریه (FT-IR) و مرئی-فرابنفش (UV-vis) استفاده شد. کوپلیمر PLA-HA از طریق کونژوگاسیون NH2-HA و آکریلات-PLA سنتز گردید و نانوذرات PLA/HA/Fe3O4/5-FU و PLA/Fe3O4/5-FU به روش انتشار حلال تهیه شدند. سایز و پتانسیل زتای نانوذرات به روش پراکندگی نور دینامیکی (DLS) تعیین گردید. جهت بررسی خصوصیات مورفولوژی و شکل نانوذرات، تصویربرداری از آنها توسط میکروسکوپ الکترونی عبوری (TEM) صورت گرفت. همچنین، خصوصیات مغناطیسی نانوذرات از طریق آنالیز مغناطیس سنجی ارتعاشی (VSM) ارزیابی گردید. کارآیی احتباس دارو در نانوذرات نیز از طریق اندازهگیری میزان داروی آزاد موجود در محلول رویی نهایی فرآیند سنتز، با روش اسپکتروفوتومتری تعیین گردید. بررسی الگوی رهش دارو از نانوذرات، از طریق انکوباسیون نانوذرات در بافر فسفات (PBS) با دو pH اسیدی (pH=6) و خنثی (pH=7.4) صورت گرفت. در نهایت، سمیت و کارآیی نانوحاملهای تهیه شده در انتقال دارو به ردهی سلولی سرطان کولورکتال (HCT116)، به ترتیب از طریق تیمار سلولها با غلظتهای متفاوت نانوذرات فاقد دارو و نانوذرات حاوی دارو و انجام تست MTT بررسی شد. نتایج و بحث: سنتز نانوذرات اکسید آهن توسط طیفهای FT-IR و UV-vis تایید گردید. نانوذرات PLA-HA/Fe3O4/5-FU سنتز شده، دارای شکل کروی، میانگین اندازهی 235 نانومتر و پتانسیل زتای -18 میلیولت بودند. نتایچ آنالیز VSM نشان داد که نانوذرات، دارای خاصیت سوپرپارامغناطیسی هستند. نتایج آنالیز اسپکتروفوتومتری حاکی از کارآیی 42 درصدی احتباس داروی 5-FU در نانوذرات بود. نانوذرات در هر دو pH اسیدی و خنثی، از الگوی رهش انفجاری و پیوسته تبعیت مینمایند. همچنین، نتایج حاکی از رهش بیشتر دارو در محیط اسیدی بود که با توجه به pH پایینتر بافت تومور نسبت به بافت سالم، این خاصیت ممکن است به کاهش اثرات جانبی دارو منجر گردد. نتایج تست MTT نشاندهندهی زیستسازگاری مناسب نانوحاملهای فاقد دارو و سمیت پایین آنها بوده است. همچنین، مشاهده گردید که نانوذرات PLA-HA/Fe3O4/5-FU در مقایسه با نانوذرات PLA/Fe3O4/5-FU سبب کاهش بیشتری در زندهمانی سلولهای HCT116 میشوند. علت احتمالی این پدیده، برهمکنش اختصاصی هیالورونیک اسید با گیرندههای CD44 با بیان بیش از حد در سطح سلولهای سرطانی است. بنابر خصوصیات قابل قبولی که در نتایج ذکر شد، این نانوذرات قابلیت پژوهش بیشتر جهت به کارگیری به عنوان یک سیستم دارورسانی هدفمند را دارند.

Evaluation of loading capacity, release profile, delivery efficiency and toxicity of 5-fluorouracil-loaded PLA-HA/Fe3O4 nanoparticles

Introduction: 5-Fluorouracil is the main drug used in colorectal cancer treatment. Due to its non-specific effect on healthy cells, 5-FU is associated with multiple side effects. A possible solution to overcome the adverse side effects of the drug, is to load the drug in targeted nanoparticles. In the present study, iron oxide (Fe3O4) nanoparticles with polylactic acid-hyaluronic acid copolymer (PLA-HA) coating were synthesized to encapsulate 5-flourouracil. Physicochemical properties, drug release pattern, toxicity and efficiency of nanoparticles in drug delivery to colorectal cancer cell lines (HCT116) were then evaluated. Methods: Fe3O4 nanoparticles were prepared by green biosynthesis method. Fourier transform infrared spectroscopy (FT-IR) and Ultraviolet–visible spectroscopy (UV-Vis) was used to confirm the synthesis of iron oxide nanoparticles. PLA-HA copolymer was synthesized by conjugation of NH2-HA and acrylate-PLA. PLA-HA/Fe3O4/5-FU and PLA/Fe3O4/5-FU nanoparticles were prepared by solvent diffusion method. The size and zeta potential of nanoparticles were determined by dynamic light scattering (DLS) method. Morphological properties and shape of the nanoparticles were studied using transmission electron microscopy (TEM) imaging. Also, the magnetic properties of the nanoparticles were evaluated by vibrational sample magnetometer (VSM). The encapsulation efficiency of 5-FU in nanoparticles was also determined through measuring the amount of free drug present in the final supernatant of the synthesis process by spectrophotometry. The drug release pattern was investigated by incubating nanoparticles in phosphate buffer solution (PBS) with acidic and neutral pH (respectively, pH 6 and 7.4). Finally, the toxicity and efficacy of prepared nanocarriers in drug delivery to colorectal cancer cell line (HCT116) were evaluated using MTT assay on cells treated with different concentrations of drug-free nanoparticles and drug-loaded nanoparticles. Results and discussion: The synthesis of iron oxide nanoparticles was confirmed by FT-IR and UV-vis spectra. The synthesized PLA-HA/Fe3O4/5-FU nanoparticles had a spherical shape with an average size of 235 nm and a zeta potential of -18 mV. The results of VSM analysis demonstrated that the nanoparticles had superparamagnetic properties. The results of spectrophotometric analysis showed that the encapsulation efficiency of 5-FU in nanoparticles was 42 percent. The nanoparticles followed a burst and sustained release pattern at neutral and acidic pH. Also, it was found that the drug is released more readily at acidic pH. Due to the lower pH of tumor tissue compared to healthy tissue, this property may reduce the side effects of the drug. MTT assay results indicate proper biocompatibility of drug-free nanocarriers and their low toxicity. It was also observed that PLA-HA/Fe3O4/5-FU nanoparticles significantly reduced the HCT116 cells viability compared to PLA/Fe3O4/5-FU nanoparticles. This phenomenon is probably due to the specific interaction of hyaluronic acid with CD44 receptors, which are overexpressed on the surface of cancer cells. Due to its acceptable properties, PLA-HA/Fe3O4 nanoparticles have the potential for further research to be used as a targeted drug delivery system.
