CTLA-4 over-expression in intracellular compartments as well as on the cell surface of lymphocytes derived from patients with lung cancer

English Abstract

Background and Objectives: CTLA-4 behave as a negative regulator and inhibit the proliferation and effective immune responses also they prevent autoimmune reactions by suppression of self- reactive T cells . recent studies have shown that the function of these immune regulators play a pivotal role in suppression and inhibition of the effective immune responses against cancer therefor¸ they are causes to tumor progression in patients with different malignancies. The purpose of the this investigation was to study lymphocytes have expressed surface and intracellular CTLA-4 in peripheral blood from non small cell lung cancer patients. Methodes: Twenty three new cases with NSCLC who received no prior treatment as well as sixteen sex-age matched healthy donors with no family history of malignancy and/or autoimmune disease were recruited. Mono nuclear cells were isolated by Ficoll-Hypaque density gradient centrifuge. Intracellular and surface staining of CTLA4 was done by monoclonal antibodies and analyzed by flow cytometric analysis was used to evaluate the prevalence of the targeted cells. Results: In both groups CTLA4 presence in intracellular compartments of lymphocytes was significantly higher than that expressed on the surface (p<0.05). Compared to the controls the expression of intracellular as well as surface CTLA4 was independently increased in lymphocytes of the patients. Total T cells from the patients had significantly higher CTLA4 expression both in intracellular compartments and on the surface. (Intracellular: 19.2±17 vs 7.32±12.2, Surface: 0.71±0.56 vs 0.46±0.19¸ p<0.05).Conclusion: our data suggest that CTLA-4 and T reg cells as a negative regulator increased in the NSCLC patients and these molecules can be one of the main causes of tumor progression and uneffective immune responses. Conclusion: our data suggest that the use of CTLA-4- targeted immunomodulatory therapy may be an effective strategy to treat NSCLC patients.
