CYTOTOXICITY OF ECBALLIUM ELATRIUM FRUIT EXTRACT ON ESOPHAGEAL (KYSE-30), GASTRIC (AGS) AND LIVER (HEPG2) CANCER CELL LINES

Background: Esophageal cancer is a relatively rare form of cancer, but some world areas have a markedly higher incidence than others: China, Iceland, India and Japan, as well as the United Kingdom, appear to have a higher incidence, as well as the region around the Caspian Sea. There are various subtype primerly adenocarcinoma (approx 50-80% of all esophageal cancer) and squamous cell cancer. Liver cancer is the sixth most common cancer worldwide. Liver cancer accounts for 5.6% of all cancer in human. Liver cancer is a significant cause of morbidity and mortality worldwide. The hepatocellular carcinoma (HLL) is the most common malignant primary liver cancer, which represent 75-90% of liver cancers worldwide. Gastric cancer is a major public health issue as the fourth most common cancer and the second leading cause of death worldwide. Gastric cancer is particularly common in Asia, Eastern Europe, and in South America. The gastric adenocarcinoma is the most common subtype of gastric cancer. In Iran especially Ardabil province, Esophageal and Gastric cancers is the most frequently diagnosed form of cancer. Ecballium Elaterium also called the Squirting cucumber or exploding cucumber is a medicinal plant in the cucumber family. It is native to Europe, northern Africa, and temperate areas of Asia. Ecballium Elaterium has been used as a medical plant. The juice of the fruit is antirheumatic, cardiac and purgative. The plant used internally in the treatment of oedema associated with kidney complaints, heart problems, rheumatism, paralysis and shingles. Externally it has been used to treat sinusitis and painful joints. Because of curative properties of this plant we investigated cytotoxic effects of this plant on these cancers. Methods andMaterials: The immature fruits were collected from rural areas of Ardabil province on August 2008, cleaned and cut into small pieces; they were crushed using homogenizer to get homogenous mixture, then filtered and centrifuged and finally freeze-dried and kept at -20C. HepG2 and AGS (liver and gastric cancer respectively) cell lines plated in 96-well plates with the concentration 2x104 (RPMI1640 90% + FBS 10%) and KYSE-30 cell line also plated with the concentration 2x104 (RPMI45%+HAM45%+FBS10%) when cells ready, plates were treated with initial concentration of 2mg/ml extract with ¼serial dilutions and incubated for 24, 48 and 72 hrs. cytotoxicity of aqueous extract of fruit was examined by three different methods, 1.MTT method 2. neutral red uptake method 3, FRAME method. Result: fruit aqueous extract of Ecballium Elatrium was showed cytotoxic effect on all cell lines at 24, 48 and 72 hrs incubation times. The cytotoxic effect was observed in different incubation times. (With longer incubation time the cytotoxicity effects augmented). Conclusion: the aqueous extract of Ecballium fruit has cytotoxic effect on KYSE-30, AGS and HepG2 cell lines.
