بررسی ارتباط بین سرطان معده و پلی مرفیسم ژن آنزیم مبدل آنژیوتانسین (ACE) 1 درافراد مبتلا به سرطان معده در استان اردبیل

Persian Abstract

سابقه و هدف: سرطان معده سومین بدخیمی شایع در ایران است که حدود %50 از سرطان های دستگاه گوارش و %55 از سرطان های منجر به مرگ در ایران را شامل می شود. سرطان معده ی استان اردبیل در جهان از شایع ترین سرطان های دنیاست. سرطان های دستگاه گوارش فوقانی، شامل بیش از %50 مرگ و میرهای سرطان در این منطقه است. ژن ACE (آنزیم مبدل آنژیوتانسین)، نقش مهمی نه تنها در تنظیم هموستاز عروقی ایفا می کند بلکه در تحریک هماتوپوئزیتر نقش ایفا می کند. پلی مرنسیم Insertion / Deletion (حذف / اضافه) در ژن ACE ، امروزه با پاتوژنز و پیشرفت سرطان های انسانی مربوط شده است. ما می خواهیم در این طرح، ارتباط بین Insertion / Deletion و پلی مرفیسم ژن ACE و استعداد به سرطان معده را در استان اردبیل بررسی کنیم. مواد و روش ها: ما 100 بیمار مبتلا به سرطان معده را با 100 فرد سالم که از نظر جنس و سن match شده بودند، انتخاب کردیم. این پلی مرفیسم طولی (length polymorphism)، با روش PCR معمولی روی DNA استخراج شده از ذهن محیطی، بررسی شد. یافته ها: در بین موارد بیماری، %19 هموزیگوت برای II ، %26 هموزیگوت برای DD و بقیه %55 ID بودند. نتایج اطلاعات کنترل %20 هموزیگوت برای II ، %46 هموزیگوت برای DD و %34 هموزیگوت برای ID بودند. هیچ ارتباط معنی داری بین موارد بیماری و کنترل ها یافت نشد (p=0/47) همچنین هیچ ارتباط پاتولوژیکی بین موارد بیماری پیدا نشد. نتایج: پیدانکردن هیچ ارتباطی در این تحقیق، می تواند ناشی از ارتباط بین پلی مرفیسم ذکر شده و بعضی بیماری ها مانند دیابت که در بعضی از بیماران تشخیص داده نشده است، باشد. اما از لحاظ تئوری، این پلی مرفیسم طولی به هدف می رسد که در استعداد به سرطان معده نقش دارد.

English Abstract

Introduction: Gastric cancer as the 3rd most common malignancy in Iran, accounts for ~50% of all GI cancers who cause 55% of all cancer-related deaths in Iran. The rates of gastric cancer reported from Ardabil province, Iran, are among the highest in the world. Upper gastrointestinal cancer accounts for more than 50% of all cancer deaths in this area. The angiotensin-converting enzyme (ACE) plays an important role not only in the regulation of vascular homeostasis but also in stimulation of hematopoiesis. The insertion/deletion polymorphism of ACE gene has recently been linked to the pathogenesis and progression of human cancers. We aimed to evaluate the association between insertion/deletion (I/D) polymorphism of the ACE gene and susceptibility to gastric cancer in our province. Materials and Methods: We enrolled 100 patients with gastric cancer and 100 age- and sex-matched healthy control participants. This length polymorphism was revealed by conventional PCR method on DNA extracted from peripheral blood. Results: Among cases, there were 19% homozygous for II, 26% homozygous for DD, and the remaining 55% were ID. The resulted Controls data were 20% homozygous for II, 46% homozygous for DD, and 34% for ID. Any significant correlations were not found between cases and controls (p=0.47) or pathologic values in case group. Conclusion: Finding any correlation in this investigation could results form the reported relationships between the noted polymorphism and some disorders such as Diabetes which were undetected in some of our participants. But, Theoretically this length polymorphism seems to offer the susceptibility to Gastric cancer.
