چند ریختی P53-ARG72PRO استعداد ابتلا به سرطان معده را در اردبیل اعطا نمی کند

Persian Abstract

زمینه: سرطان معده به عنوان سومین بدخیمی شایع در ایران، مسئول حدود 50 درصد از کل سرطانهای دستگاه گوارش است که باعث حدود 55٪ از مرگ های مرتبط با سرطان در ایران می شود. نرخ گزارش شده سرطان معده در استان اردبیل در میان بالاترین موارد در سطح جهان می باشد. چند ریختی کدون 72 ژن TP53با نمو سرطانهای گوناگونی در نقاط مختلف جهان ارتباط داده شده است. چند ریختی منجر به تبدیل اسید امینه آرژی نین (با رمز CGC) به پرولین ( با رمز CCC) می شود. هدف ما بررسی ارتباط این چند ریختی با ابتلا به سرطان معده در استان اردبیل بود. روشها: 100 بیمار با میانگین سنی 65.9 سال مبتلا به سرطان معده اولیه و گروه سالم مشابه از نظر سنی و جنسی به عنوان گروه کنترل تحت مطالعه قرار گرفتند. بررسی با روش PCR-RFLP بر رروی DNA استخراج شده از خون محیطی افراد شرکت کننده انجام پذیرفت. نتایج: در گروه بیماران ژنوتیپهای Arg/Arg، Arg/Proو Pro/Pro به ترتیب 16.1٪ ، 42.5٪ و 41.4٪ بود. حال آنکه این اعداد برای گروه کنترل به ترتیب 18.5٪ ، 40.2٪ و 41.4٪ بود. در مقایسه گروه بیماران و کنترل، اختلاف معنی داری مشاهده نگردید. بحث: به علت فراوانی بالای سرطان معده در استان اردبیل، مطالعه نقش عوامل مستعد کننده ژنتیکی سرطان معده بسیار حائز اهمیت می باشد. علیرغم یافتن هیچگونه ارتباط معنی دار میان چند ریختی مذکور، بررسی عوامل ژنتیکی دیگر توصیه می گردد.

P53-ARG72PRO POLYMORPHISM CONFERS ANY SUSCEPTIBILITY TO GASTRIC CANCER IN ARDABIL

English Abstract

Background: Gastric Cancer (GC) as the third most common malignancy in Iran, accounts for �50% of all GI cancers who cause 55% of all cancer-related deaths in Iran. The rates of GC reported from Ardabil Province are among the highest in the world. Upper gastrointestinal cancer accounts for more than 50% of all cancer deaths in this area. Codon 72 polymorphism of the tumor suppressor gene TP53 has been associated with a higher risk in the development of several types of cancer. The polymorphism results in a variant protein with either an arginine (CGC) or a proline residue (CCC). We aimed analyze the association of the TP53 codon 72 polymorphism with the risk of developing gastric cancer in a high-risk population around the world. Methods: We enrolled 100 patients with mean age 65.9 Yrs. affected with primary GC and same age- and sex- matched healthy control participants. The analysis has been carried out by PCR-RFLP on DNA extractions from peripheral blood. Results: In the case group, the genotype was 16.1%, 42.5%, and 41.4% for Arg/Arg, Arg/Pro, and Pro/Pro, respectively. And for controls were 18.5%, 40.2%, and 41.4%. In comparing case and control group, any significant difference has been found. Conclusion: Because of high frequency of GC in our province, the investigations about the role of genetic susceptibilities for GC are very important. In spite of finding no relationship between P53 polymorphisms, studying other genetic variations is recommended.
