تعیین حساسیت و اختصاصیت روش PCR در تشخیص بروسلوز انسانی

Persian Abstract

زمینه و هدف : بروسلوز یکی از شایع ترین بیماري هاي مشترك بین انسا ن و دام در ایران می باشد . در اغلب موارد تشخیص بروسلوز دشوار است و دلیل این دشواري نه فقط به خاطر شباهت بالینی این بیماري با بسیاري از بیماري هاي عفونی و غیرعفونی می باشد بلکه هم چنین روش هاي تشخیصی در اغلب موارد موفق به جداسازي ارگانیسم نمی گردند . در PCR می باشد. هدف این مطالعه تعیین حساسیت و ویژگی روش PCR یکی از روش هاي تشخیصی سریع و ایمن تشخیص بروسلوز انسانی با استفاده از نمونه هاي سرم می باشد. مواد و روش ها: این مطالعه از نوع ارزیابی تست هاي تشخیصی می باشد. افراد شرکت کننده در این مطالعه شامل 30 فرد داراي علائم بالینی و تست رایت مثبت که گروه بیماران ما را تشکیل می دادند و 30 فرد سالم و با تست رایت منفی که گروه شاهد ما بودند. 30 نمونه سرم از افراد گروه بیماران و 30 نمونه سرم از افراد سالم مورد استفاده قرار گرفت. بر انجام شد. PCR روي این نمونه ها آزمایش مثبت PCR مثبت شدند و از افراد سالم، 4 مورد با روش PCR یافته ها: از 30 فرد گروه بیماران، 15 نمونه با روش 86 درصد تعیین شد. / 50 درصد و ویژگی آن 6 PCR شدند. حساسیت روش نتیجه گیري: هر چند در تعدادي از مطالعات نمونه ارجح براي تشخیص بروسلوز را سرم می دانند، ما به حسا سیت و با استفاده از نمونه سرم جهت تشخیص بروسلوز دست نیافت ه ایم . بدین منظور، PCR اختصاصیت بالایی براي روش استفاده از ترکیبی از روش هاي آزمایشگاهی را براي تشخیص بروسلوز انسانی پیشنهاد می کنیم.

Determination of sensitivity and specificity of PCR in diagnosis of human brucellosis

English Abstract

Background: Brucellosis is one of the most common zoonotic diseases in Iran. In most cases, the diagnosis of brucellosis is difficult not only because of its clinical similarity to many infectious and noninfectious diseases, but also because diagnostic methods often fail to detect organisms. PCR is a rapid and safe diagnostic method applied to the diagnosis of brucellosis. The purpose of this study was to determine the sensitivity and specificity of PCR for diagnosis of human brucellosis by using serum samples. Materials and Methods: This study which was done to evaluate diagnostic tests included30 serum samples from patients with clinical presentation of brucellosis with positive Wright test and serum samples of30 healthy people with negative Wright test. These samples were examined by PCR. Results: PCR results were positive for 15 samples of the patients group in comparison with 4 samples from the 30 healthy subjects. The sensitivity and specificity of PCR were 50% and 86.6%, respectively. Conclusion: Although in some studies, the preferred sample for diagnosis of brucellosis was serum, in this study, PCR on serum samples did not indicate high sensitivity and specificity in diagnosis of brucellosis. Hence, using a combination of methods for diagnosis of human brucellosisis suggested.
