بررسی نسبت آنزیمهای eNOS و MMP-2 در افراد مبتلا به سرطان معده

سابقه و هدف: آنزیمهای نیتریک اکسید سنتاز و ماتریکس متالوپروتئیناز به ترتیب با سنتز نیتریک اکسید و پروتئولیز بسیاری از ترکیبات ماتریکس خارج سلولی، نقش مهمی را در القای سرطان و متاستاز نشان میدهند. مطالعه ای در سطح سلولی نشانگر این است که القا بیان نیتریک اکسید سنتاز موجب کاهش فعالیت MMP-2 میشود. هدف اصلی مطالعه حاضر بررسی نسبت آنزیم های اکسید نیتریک سنتاز و ماتریکس متالوپروتئینازها در افراد سالم و مبتلا به سرطان معده و مقایسه آنها با هم بود. مواد و روشها: در این مطالعه مورد- شاهدی تعداد 50 نفر بیمار مبتلا به سرطان معده مراجعه کننده به کلینیک ارس به صورت تصادفی و 30 نفر فرد سالم با جنس و سن مشابه انتخاب شدند. پس از خونگیری و جداسازی سرم، میزان آنزیمهای نیتریک اکسید سنتاز اندوتلیالی و ماتریکس متالوپروتئیناز 2 با روش الیزا (ELISA) سنجش گردید. یافته ها: مطالعه حاضر نشان میدهد که نسبت آنزیم MMP-2/eNOS در افراد بیمار به طور معنی داری بیش از افراد سالم است، در حالی که eNOS مبتلایان به سرطان معده به طور معنی داری کمتر از افراد سالم است و MMP-2 تغییر معنی داری را نشان نمی دهد. نتیجه گیری: برخلاف مطالعات قبلی در سطح سلولی، مطالعه حاضر نشان می دهد که سطح سرمی eNOS و MMP-2 باهم ارتباطی ندارند. بنابراین ممکن است عوامل دیگری در تنظیم میزان MMP-2 سرمی غیر از eNOS نقش داشته باشند.

Evaluation of eNOS and MMP-2 enzymes in patients with gastric cancer

English Abstract

Background & objects: Nitric oxid synthase and matrix metalloproteinase enzyme plays a important role in cancer and metastasis inducing with constriction of extracellular matrix.previous study in cellular level indicates, incluetion of expression eNOS decrease MMP-2 activity. Main object of the present study was checking of eNOS/MMP-2 in the healthy subject and patients with Gastric cancer and compared them together. Material and methods: in this case-control study 50 patients whit gastric cancer, that are refered to ARAS clinic, randomly, also, 30 healthy individuals with similar age and sex selected.after separation of blood, serum enzyme level of nitric oxide synthase and MMP-2 were measured by ELISA. Results: The present study show that MMP-2/eNOS enzyme in the patients is significantly more than healthy individual. Wherease eNOS level in the gastric cancer patients is significantly less than healthy individuals, But MMP-2 enzyme dosen't show significant differentiation in two groups. Conclusions: contrary to previousstudy in the cellular level, this study shows serum level of eNOS & MMP-2 don't have relation together. Thus, propably there are other factors that play a role in the regulation of MMP-2 level, beyond eNOS
