بررسی بیان ژن BRCA1 در بیماران مبتلا به سرطان پستان در استان اردبیل

Persian Abstract

زمینه و هدف: سرطان پستان از جمله شایعترین بیماریهاي زنان در سراسر جهان می باشد. ژن 1 ژن سرکوبگر تومور شناخته شده است که فقدان آن منجر به آسیبهاي متنوعی در ژنوم می گردد. بنابراین تغییرات در می تواند منجر به آسیب پذیري سلول پستانی نسبت به عملکرد انکوژنها گردد. در این مطالعه BRCA سطح بیان ژن 1 و داده هاي پاتولوژیکی تومورهاي سرطان پستان، علی الخصوص BRCA سعی بر بررسی ارتباط میان سطح بیان ژن 1 درجه تومور می باشد. RT-PCR در 27 بیمار مبتلا به سرطان پستان اولیه از روشهاي BRCA روش کار: جهت بررسی کیفی و کمی بیان ژن 1 جهت جلوگیري از کسب پاسخهاي منفی کاذب) ) Nested RT- PCR استفاده شد و بهره گیري از روش Q-RT-PCR و به افزایش دقت آزمایش یاري رساند. BRCA را نشان دادند. ارتباطات معنی دار میان وجود بیان 1 BRCA یافته ها: هشتاد و پنج درصد از بیماران، بیان ژن 1 (P< و درجه تومور بیماران نیز ارتباط معنی دار ( 0.05 BRCA و درجه تومور مشاهده گردید. میان سطح بیان ژن 1 بدست آمد. به گونه اي که کاهش بیان ژن با افزایش درجه تومور مرتبط بود. در سرکوب پیشرفت تومور از راههاي مختلفی نظیر ترمیم BRCA نتیجه گیري: با توجه به نقش پر اهمیت ژن 1 با BRCA آسیب دیده و همسو با دیگر نتایج بدست آمده، مطالعه حاضر آشکار ساخت که کاهش بیان ژن 1 DNA افزایش وخامت تومور مرتبط است.

Investigation the BRCA1 gene expression level in patients affected with breast cancer in Ardabil province

English Abstract

Background & Objectives: Breast cancer is among the most frequent disorders among the world. BRCA1 gene was known as a tumor suppressor gene which its loss could be resulted in various destructions in genome. Therefore, changes occurred in its expression level could lead to increasing the effects of oncogene activity on breast cells. Methods: RT-PCR and Q-RT-PCR were used for detecting BRCA1 expression status among 27 primary affected breast cancer patients. The association between its expression status and histopathologic data had been evaluated by statistical assays. Results: Eighty-five percent of patients showed BRCA1 expression and this expression as well as its quantity had significant associations with tumor grade. It was found that tumors with low levels of BRCA1 expression tend to have higher grades. Conclusion: BRCA1 has an important role in suppressing breast tumorogenesis. Corresponding to some of the previous reports, our findings revealed the role of decreasing in BRCA1 expression level as one of the factors which allows tumors to progress
