SYBR Green based RT-TRAP assay of the Telomerase activity in patients affected with gastric or esophageal cancers in Ardabil province
English Abstract
Background and objective : astric cancer is the fourth most common cancer and second leading cause of cancer related death in the world with under 5-year survival . Original Article Gastric and esophageal cancers are the most frequent causes of cancer-related deaths in Iran with the highest rates reported from Ardebil.The survival incidence for patients affected with these cancers in ardebil is under 2 years. Telomerase is a cellular reverse transcriptase (molecular motor) that adds new DNA onto the telomeres that are located at the ends of chromosomes. Telomerase is repressed in most human somatic cells, but reactivated in more than 80% of all human cancers. Materials and methods : 30 patient with Gastric and Esophageal Cancer that their disease confirmed histologically from ardabil emam khomaine hospital were studied in this project. Then telomerase protein was extracted by chaps buffer and Protein concentration of extracts was determined with the Protein Assay (Bio-Rad), Then telomerase activity assaied with both TRAP Assay and SYBR Green RT-TRAP Assay. Results : 90% of Patients Affected with Gastric and Esophageal Cancer had telomerase activity. Discussion : telomerase is potent biomarker for diagnosis and cancer therapotic.
