ﺑﺮرﺳﯽ ازدﯾﺎد ﻧﺴﺦ ژن hTERT ﺑﻪ ﻋﻨﻮان ﻧﺸﺎﻧﮕﺮ ﭘﯿﺶ آﮔﻬﯽ در ﺑﯿﻤﺎران ﻣﺒﺘﻼ ﺑﻪ ﺳﺮﻃﺎن ﭘﺴﺘﺎن در اﺳﺘﺎن اردﺑﯿﻞ

ﺳﺮﻃﺎن ﭘﺴﺘﺎن ﺷﺎﯾﻌﺘﺮﯾﻦ ﺳﺮﻃﺎن در زﻧﺎن ﺟﻬﺎن ﺑﻮده و ﯾﮏ ﺳﻮم ﺳﺮﻃﺎن ﻫﺎ را ﺗﺸﮑﯿﻞ ﻣﯽ دﻫﺪ. آﻧﺰﯾﻢ ﺗﻠﻮﻣﺮاز ﯾﮏ رﯾﺒﻮ ﻧﻮﮐﻠﺌﻮﭘﺮوﺗﺌﯿﻦ ﺑﺎ ﻓﻌﺎﻟﯿﺖ ﺗﺮاﻧﺲ ﮐﺮﯾﭙﺘـﺎز ﻣﻌﮑـﻮس ﻣـﯽ ﺑﺎﺷـﺪ ﮐﻪ ﺗﻮاﻟﯿﻬﺎي ﺗﮑﺮاري TTAGGG را ﺑﻪ اﻧﺘﻬﺎي ﮐﺮوﻣﻮزوﻣﻬﺎ اﺿﺎﻓﻪ ﻣﯽ ﮐﻨﺪ. ﺗﮑﺜﯿﺮ زﯾﺮ واﺣﺪ ﮐﺎﺗﺎﻟﯿﺘﯿﮑﯽ اﯾﻦ آﻧـﺰﯾ ﻢ (hTERT) در رده ي ﺳـﻠﻮل ﺳـﺮﻃﺎﻧﯽ و ﺗﻮﻣﻮرﻫـﺎي ﻣﺨﺘﻠـﻒ اﻧﺴﺎﻧﯽ ﻧﺸﺎن داده ﺷﺪه اﺳﺖ. ﺑﻨﺎﺑﺮ اﯾﻦ در ﭘﮋوﻫﺶ ﺣﺎﺿﺮ، ازدﯾﺎد ﻧﺴﺦ ژن hTERT در ﺳﺮﻃﺎن ﭘﺴﺘﺎن اوﻟﯿﻪ ﻣﻮرد ﻣﻄﺎﻟﻌﻪ ﻗﺮار ﮔﺮﻓﺖ. ﻣﻮاد وروﺷـﻬﺎ: 50 ﻧﻤﻮﻧـﻪ ﺗﻮﻣـﻮر ﭘﺎراﻓﯿﻨـﻪ ي ﺑﯿﻤﺎران ﻣﺒﺘﻼ ﺑﻪ ﺳﺮﻃﺎن ﭘﺴﺘﺎن و 50 ﻧﻤﻮﻧﻪ ﺑﺎﻓﺖ ﭘﺎراﻓﯿﻨﻪ ي ﺑﯿﻤﺎران ﻏﯿﺮ ﻣﺒﺘﻼ، از ﺑﺨﺶ ﭘﺎﺗﻮﻟﻮژي ﺑﯿﻤﺎرﺳﺘﺎن اﻣﺎم ﺧﻤﯿﻨﯽ (ره) اﺳﺘﺎن اردﺑﯿﻞ ﺗﻬﯿﻪ ﺷﺪ و DNA ﻧﻤﻮﻧﻪ ﻫـﺎ ﺑـﺎ روش دﺳـﺘﯽ ﻓﻨﻞ ﮐﻠﺮوﻓﺮم اﺳﺘﺨﺮاج ﮔﺮدﯾﺪ، ﺳﭙﺲ ازدﯾﺎد ژن ﻣﻮرد ﻧﻈﺮ ﺑﺎ روش ﮐﻤﯽ Real time) PCR) ﻣﻮرد ﺑﺮرﺳﯽ ﻗﺮار ﮔﺮﻓﺖ. ﯾﺎﻓﺘﻪ ﻫـﺎ: ﻣﻘﺎﯾﺴـﻪ ﻧﺘـﺎﯾﺞ ﺣﺎﺻـﻞ از دو ﮔـﺮوه ﻣـﻮرد ﻣﻄﺎﻟﻌﻪ، ازدﯾﺎد ﻧﺴﺦ ژن hTERT را 2/71 ﺑﺮاﺑﺮ ﻧﺸﺎن داد. ﺑﺤﺚ و ﻧﺘﯿﺠﻪ ﮔﯿﺮي: اﯾﻦ ﻣﻄﺎﻟﻌـﻪ ﻧﻈﯿـﺮ اﮐﺜـﺮ ﻣﻄﺎﻟﻌـﺎت ، اﻓـﺰاﯾﺶ ازدﯾـﺎد ﻧﺴـﺦ ژن hTERT را در ﺑﯿﻤـﺎران ﻣﺒـﺘﻼ ﺑـﻪ ﺳﺮﻃﺎن ﭘﺴﺘﺎن ﻧﺴﺒﺖ ﺑﻪ ﮔﺮوه ﮐﻨﺘﺮل ﻧﺸﺎن داد، ﮐﻪ ﻣﯽ ﺗﻮاﻧﺪ ﺑﻪ ﻋﻨﻮان ﭘﯿﺶ آﮔﻬﯽ در ﺑﯿﻤﺎران ﻣﺒﺘﻼ ﺑﻪ ﺳﺮﻃﺎن اوﻟﯿﻪ در اﺳﺘﺎن اردﺑﯿﻞ ﻣﻄﺮح ﺑﺎﺷﺪ.

Investigation of the hTERT Amplification as a Prgnostic Marker in Patients Offected with Breast in Ardabil

English Abstract

Breast cancer is the most common cancer in the world’s women and makes up one- third of cancers. Telomerase enzyme is a ribonucleoprotein with reverse transcriptase activity that adds TTAGGG repeated sequences to the ends of the chromosomes. Reproduction of the catalytic subunit of the enzyme (hTERT) in cancer cell lines and different human tumors is shown. So, in tht present investigation, the amplification of the hTERT gene in primary breast cancer was studied. Materials and Methods : Fifty paraffin-embedded tumor samples from breast cancer patients and 50 paraffin tissue samples from healthy patients was provided from Ardabil Imam Khomeini Hospital’s Pathology section and DNA samples were manually extracted with phenol-chloroform based method. Then the amplification of the desired gene was evaluated by quantiative (Real- Time) PCR. Results: Comparing the results of the two geoups revealed 2.71 fold increasing of the hTERT gene copy number. Discussion and conclusion: Corresponding many studies, our result indicated the increased amplification of hTERT gene in breast cancer patients compared to the control group which could be considered as prognosis factor for the primary breast cancer patients in Ardabil.
