Immunogenicity of a DNA Vaccine Encoding Ag85a-Tb10.4 Antigens from Mycobacterium Tuberculosis

خلاصه انگلیسی

Background and Objectives:Tuberculosis is a life threatening disease that nowadays prevented by BCG vaccine. Development more effectual vaccines than BCG is the urgent priority in TB control. Ag85a and Tb10.4 is the member of culture filter protein (CFP) of M. tuberculosis that has high immunogenicity. The aim of this study was to analyze immunogenicity of Ag85a-Tb10.4 DNA vaccine by enzyme-linked immunosorbent (ELISA) assay.Materials and methods:In this study a plasmid DNA vaccine encoding Ag85a-Tb10.4 that constructed in previous study was used to examine its capability in stimulation immune responses in animal model. Female BALB/c mice were vaccinated with 100μg of purified recombinant vector and level of five cytokines including IFN-γ, IL-12, IL-4, IL-10 and TGF-β were measured. Result:In this study the immunogenicity of the recombinant vector was tested in BALB/c mice. Amount of IFN-γ and IL-12 for the mice following immunization with Ag85A-Tb10.4 were significantly greater than the BCG and control group. In other cytokines Between BCG and recombinant vaccine there were not significant differences. Conclusion:A DNA vaccine encoding Ag85A-TB10.4 from M. tuberculosis was administered to BALB/c mice by intramuscular needle injection and its effectiveness was determined.IFN-γ and IL-12 as Th1 marker increased significantly in mice vaccinated with Ag85a-TB10.4 DNA vaccine in compare to control and BCG groups. Our results may serve as groundwork for further research into the prevention and treatment of tuberculosis disease.
