Persian Abstract

ﭼﮑﯿﺪه زﻣﯿﻨﻪ و ﻫﺪف: ﺑﯿﻤﺎري ﻣﺎﻟﺘﯿﭙﻞ اﺳﮑﻠﺮوزﯾﺲ ﯾﮏ ﺑﯿﻤﺎري ﻣﺰﻣﻦ دﺳﺘﮕﺎه ﻋﺼﺒﯽ ﻣﺮﮐﺰي اﺳﺖ. از آن ﺟﺎﯾﯽ ﮐﻪ ﺳﻄﺢ ﻫﻮرﻣﻮن ( ﺗﺤﺖ ﺗﺄﺛﯿﺮ ﻫﻢ ﻗﺮار ﻣﯽ MS ﻫﺎي ﺟﻨﺴﯽ و ﺑﯿﻤﺎري ﻣﻮﻟﺘﯿﭙﻞ اﺳﮑﻠﺮوزﯾﺲ) ﮔﯿﺮﻧﺪ. ﻫﺪف از اﯾﻦ ﻣﻄﺎﻟﻌﻪ، ﺑﺮرﺳﯽ ﺗﺄﺛﯿﺮ ﻋﺼﺎره ﻫﯿﺪرواﻟﮑﻠﯽ ﻗﺎرچ دﻧﺒﻼن ﺑﺮ ﺳﻄﺢ ﻫﻮرﻣﻮن اﺳﺘﺮوژن و ﭘﺮوژﺳﺘﺮون در ﻣﻮش ﺻﺤﺮاﯾﯽ ﻣﺎده ﻣﺪل ام اس ﺗﺠﺮﺑﯽ اﺳﺖ. ﻣﻮاد و روش 6 ﮔﺮم در 180±10 ﺳﺮ ﻣﻮش ﺻﺤﺮاﯾﯽ ﻣﺎده ﺑﺎﻟﻎ ﻧﮋاد وﯾﺴﺘﺎر ﺑﺎ وزن ﺗﻘﺮﯾﺒﯽ 42 ، ﻫﺎ: دراﯾﻦ ﭘﮋوﻫﺶ ﺗﺠﺮﺑﯽ ﺗﺎﯾﯽ ﻣﻮرد ﻣﻄﺎﻟﻌﻪ ﻗﺮار ﮔﺮﻓﺘﻨﺪ. ﮔﺮوه ﮐﻨﺘﺮل ﻧﺮﻣﺎل ﺑﺪون ﺗﯿﻤﺎر داروﯾﯽ ﺑﻮد و ﮔﺮوه 7ﮔﺮوه 40 ﺑﻪ ﻣﺪت 5 ﺗﺎ 1 ﻫﺎي ﺗﺠﺮﺑﯽ ﻣﯿﻠﯽ330 و 220 ،110 ﻋﻼوه ﺑﺮ ﮐﻮﭘﺮﯾﺰون ﺑﻪ ﺗﺮﺗﯿﺐ ﻧﺮﻣﺎل ﺳﺎﻟﯿﻦ و 5 ﺗﺎ 2روز ﮐﻮﭘﺮﯾﺰون درﯾﺎﻓﺖ ﮐﺮدﻧﺪ. ﮔﺮوه ﺗﺠﺮﺑﯽ ﮔﺮم ﺑﺮ ﮐﯿﻠﻮﮔﺮم وزن ﺑﺪن ﻋﺼﺎره 0/2 روز ﺑﺎ ﺣﺠﻢ 12 ي ﻫﯿﺪرواﻟﮑﻠﯽ ﻗﺎرج دﻧﺒﻼن را ﺑﻪ ﺻﻮرت ﺗﺰرﯾﻖ داﺧﻞ ﺻﻔﺎﻗﯽ ﺑﻪ ﻣﺪت ﻣﯿﻠﯽ ﻟﯿﺘﺮ ﺑﻌﺪ از اﯾﺠﺎد ام اس درﯾﺎﻓﺖ ﮐﺮدﻧﺪ. در ﭘﺎﯾﺎن روز دوازدﻫﻢ از ﻫﻤﻪ ي ﮔﺮوه ﻫﺎ ﺧﻮن ﮔﯿﺮي ﺑﻪ ﻋﻤﻞ آﻣﺪه و ﻣﯿﺰان ﻫﻮرﻣﻮن ﻫﺎي ﺟﻨﺴﯽ اﻧﺪازه ﮔﯿﺮي ﮔﺮدﯾﺪ. ﯾﺎﻓﺘﻪ ﻫﺎ: ﻧﺘﺎﯾﺞ ﺑﻪ دﺳﺖ آﻣﺪه ﻧﺸﺎن داد ﮐﻪ ﮐﻮﭘﺮﯾﺰون ﺑﺎﻋﺚ ﮐﺎﻫﺶ ﺳﻄﺢ اﺳﺘﺮوژن، ﭘﺮوژﺳﺘﺮون و وزن ﻣﯽ ﺷﻮد؛ ﻫﻢ ﭼﻨﯿﻦ و ﭘﺮوژﺳﺘﺮون در 4 ﺗﺰرﯾﻖ ﻋﺼﺎره ﻫﯿﺪرواﻟﮑﻠﯽ دﻧﺒﻼن ﺑﺎﻋﺚ ﺷﺪ ﻣﯿﺰان ﻏﻠﻈﺖ ﺳﺮﻣﯽ ﻫﻮرﻣﻮن اﺳﺘﺮوژن در ﮔﺮوه ﺗﺠﺮﺑﯽ ﻧﺴﺒﺖ ﺑﻪ ﮔﺮوه ام اس اﻓﺰاﯾﺶ ﻣﻌﻨﺎ 5 و4ﮔﺮوه ﺗﺠﺮﺑﯽ داري داﺷﺘﻪ اﺳﺖ. ﻧﺘﯿﺠﻪ ﮔﯿﺮي: ﻧﺘﺎﯾﺞ اﯾﻦ ﻣﻄﺎﻟﻌﻪ ﻧﺸﺎن داد ﮐﻪ ﻋﺼﺎره ﻣﯿﻠﯽ330 و 220 ي ﻫﯿﺪرواﻟﮑﻠﯽ دﻧﺒﻼن در دوز ﮔﺮم ﺑﺮ ﮐﯿﻠﻮﮔﺮم وزن ﻣﯿﺰان اﺳﺘﺮوژن و ﭘﺮوژﺳﺘﺮون را در ﻣﻮش،ﺑﺪن ﻣﻮش ﺻﺤﺮاﯾﯽ ﻫﺎي ﻣﺪل ﺗﺠﺮﺑﯽ ﻣﻮﻟﺘﯿﭙﻞ اﺳﮑﻠﺮوزﯾﺲ اﻓﺰاﯾﺶ ﻣﯽ دﻫﺪ . واژﮔﺎن ﮐﻠﯿﺪي: ﻣﺎﻟﺘﯿﭙﻞ اﺳﮑﻠﺮوزﯾﺲ، ﻗﺎرچ دﻧﺒﻼن، اﺳﺘﺮوژن، ﭘﺮوژﺳﺘﺮون، ﻣﻮش ﺻﺤﺮاﯾﯽ


The Effect of Hydroalcoholic Extract of Truffle on Estrogen and Progesterone Levels in Experimental Model of Multiple Sclerosis (MS) in Female Rats

English Abstract

Background: The multiple sclerosis is a chronic disease of the central nervous system. Since the level of sex hormone and multiple sclerosis (MS) disease affects one another, the aim of this study was to evaluate the impact of the hydroalcoholic extract of truffle on the hormone levels of estrogen and progesterone administered in experimental model of MSinduced rats. Materials and Methods: In this experimental study, 42 Wistar female rats, weighing 180±10 grams selected into 6 groups each consisting of 7 rats. Normal control didn’t receive any treatment and experimental group was given Cuprizone toxin (as a MS model inducer) for 40 days. The experimental groups (2, 3, 4 and 5) in addition to Cuprizone received the normal saline, 110, 220 and 330 mg/kg/0.2ml (i.p.) of Hydroalcoholic extract of truffle for 12 days too. Blood samples were taken at the end of the twelfth day from all groups involved and levels of sex hormones were measured. Results: Cuprizone decreases estrogen, progesterone levels and also causes weight loss, while injection of hydroalcoholic extract of truffle increased serum levels of estrogen (in experimental group 4) and progesterone (in experimental group 4 & 5) compared to MSinduced group. Conclusion: Results of the study revealed that the hydroalcoholic extract of truffle (at dosages of 220 and 330 mg/kg) could increase estrogen and progesterone levels in rats experienced experimental multiple sclerosis.
