RELAXANT EFFECT OF THYMOQUINONE ON TRACHEAL CHAINS OF GUINEA PIG

In previous studies, the relaxant, anticholinergic (functional antagonism), and antihistaminic effects of Nigella sativa have been demonstrated on guinea pig tracheal chains. To elucidate the main substance responsible for the relaxant effect of this plant, the effect of thymoquinone, one of the main constituents of Nigella sativa was examined in this study. Therefore, in the present study the bronchodilatory effects of three cumulative volumes (0.2, 0.4, and 0.4ml) of 10 mM thymoquinone were examined by their relaxant effects on pre-contracted tracheal chains of guinea pigs by 1) 10uM methacholine and 2) 60 mM KCl and were compared with the effects of saline, theophylline, and macerated and aqueous extracts of Nigella sativa (n=5 for each group). The results showed that the relaxant effects of theophylline and extracts from Nigella sativa compared to saline in both groups if experiments (p<0.05-p<0.005) However non of the three volumes (concentration) of thymoquinone showed any relaxant effect on both groups of experiments. There were significant differences between the relaxant effect of theophylline and extracts with those of all volumes of thymoquinone (p<0.001). These results indicated that the relaxant effect of Nigella sativa is not due to its constituent thymoquinone.
